This disposable counting chamber is a single-use product with two counting chambers or 4 counting chambers (M-NZ-4CH) made of plastic with quartz properties, designed for normalised and standardised counting.
The handling is simple: 10 µl of the cell suspension are applied to the chip and counted. The coordinate system corresponds exactly to the "Neubauer Improved" counting chamber. It consists of nine squares of 1 x 1 mm each with 0.1 mm depth. Each square has a total volume of 0.1 mm³ or 10-⁴ cm³. The central square is divided into 25 small squares with triple lines and the 4 corner squares are divided into 16 small squares.
Methodology
1. General method with the C-chip
• Mix the sample well, fill the chamber with 10 µl using the capillary action
• Counting cells under the microscope
2. Mammalia cell count
• Treat cell sample with Typsin-EDTA, carefully pipette off supernatant without disturbing the pellet
• Dilute pellet with growth medium or PBS to a final concentration of 5 x 10³ cells / ml to 5 x 10⁶ cells / ml
• Load C-chip with 10 µl of the dilution, count 5 large squares
3. Red blood cell count (1:200 dilution)
• Dilute blood according to known laboratory method
• Load C-Chip with 10 µl of the dilution
• Counting the dilution on the 5 small squares (4 small corner squares and one small centre square)
• of the large centre square
4. Leukocyte count (1:20 dilution)
• Dilute blood according to known laboratory method
• C-chip loaded with 10 µl of the dilution
• Counting the dilution on the 4 large corner squares
